Development and evaluation of test methods for the detection and enumeration of opportunistic waterborne pathogens from the hospital environment

背景

緑膿菌（Pseudomonas aeruginosa）以外の多くのグラム陰性菌は水系アウトブレイクに関与しているが、これらの微生物の検査室での標準化された検出法は確立されていない。

目的

本研究の目的は、6 種類の水系病原体、すなわちアシネトバクター（Acinetobacter）属菌、バークホルデリア（Burkholderia）属菌、カプリアビダス（Cupriavidus）属菌、デルフチア・アシドボランス（Delftia acidovorans）、エリザベトキンギア（Elizabethkingia）属菌、ステノトロホモナス・マルトフィリア（Stenotrophomonas maltophilia）の検査室での検査法を確立することである。

方法

水サンプルは UK Health Security Agency の検査室で処理され、分析のためにグラスゴー王立病院（Glasgow Royal Infirmary）の検査室に送られた。水サンプルを対象微生物の純粋培養物、または環境の正常微生物叢を含有する水中の対象微生物のいずれかで処理し、その方法で混合培養から微生物を同定できるのを確認した。培地上で陰圧に保ち 100 mL の量を濾過し、30°C と37°C で培養した。培養期間は 7 日で、2 日、5 日、7 日目に培地の読み取りを行った。マトリックス支援レーザー脱離イオン化飛行時間型質量分析法（MALDI-TOF MS）を用いて、コロニーのさらなる同定を実施した。

結果

トリプティックソイ寒天培地、バシトラシン添加チョコレート寒天培地、シュードモナス選択寒天培地上での水サンプル培養により、微生物の至適回収率が得られた。分離の至適温度は 30°C であった。至適培養期間は 5 日で、すべての試験菌種が MALDI-TOF MS により確実に同定された。

結論

記載した方法は、試験した 6 種類の水系病原体を確実に検出でき、アウトブレイク調査時に水系サンプルの検査に関与する検査室で利用できる。

監訳者コメント ：

6 種類の水系病原体を定期的に環境中から検出し、また何らかの水系病原体のアウトブレイクが疑われる際にスクリーニング法として検出するのは有意義かもしれない。
