背景

手指消毒剤の殺菌効果は、通常、標準定量的浮遊試験およびフィンガーパッド試験によって評価されるが、これらの試験は生物学的リスク、高コストまたは試験に要する時間が原因で、長期持続製剤の評価ができないか、実用的でない。市販の手指消毒剤の長期持続的殺菌作用の宣伝利用が増加するのに伴い、代替となる試験戦略が必要である。

目的

アルコール非含有の手指消毒剤の長期持続効果を再現可能な方法で評価するため、標準生体外ブタ皮膚モデルの使用を調査すること。

方法

黄色ブドウ球菌(Staphylococcus aureus)、大腸菌(Escherichia coli)、肺炎桿菌(Klebsiella pneumoniae)、エンベロープウイルスである SARS-CoV-2 に対するアルコール非含有の手指消毒剤の殺菌効果を、定量的浮遊試験(EN13727、EN14476)を用いて、接触時間 5 分で試験した。次に、皮膚擦過の影響をシミュレートするため、生体外ブタ皮膚モデルを PAS 2424 の改良版と合わせて用いて、本製品を 6 時間にわたり試験した。

結果

定量的浮遊試験において、接触時間 5 分以内にすべての微生物が 5 log10 を超えて減少した。ブタ皮膚試験によって、すべての時点で低下しているものの一貫性のある有効性が示され、擦過による有効性への有意な影響はないことが示された。

結論

生体外ブタ皮膚モデルの使用は、長期持続性手指消毒剤を試験するための潜在的に実行可能で簡便なモデルシステムをもたらし、長期持続性手指消毒剤の皮膚への作用に関する宣伝利用に道筋を示すものである。

監訳者コメント


COVID-19 流行に伴い、市販の手指消毒剤の使用が増えているが、長期持続的殺菌作用の実用的な評価ができず、商業的に長期持続的殺菌作用を正確に謳うことが困難である。本研究のような生体外ブタ皮膚モデルはその評価方法として、候補となりうる。ウイルスの評価が今回は SARS-CoV-2 だったが、手指を介して伝播するノロウイルス(代用ウイルス)や別のエンベロープウイルス代用ウイルス(ワクシニアウイルスなど)を使用する方がより適切であったと考察で述べられている。
