過酸化化合物の新規殺微生物剤のin-vitroでの殺微生物活性および毒性の評価:過酸化2-ブタノン

過酸化2-ブタノン(2-butanone peroxide)のモノマー分子は、病院環境で殺微生物剤としての使用が見込まれる新規過酸化化合物誘導体である。本研究の目的は、欧州規格(European Standard;EN)ガイドラインに従い、細菌、抗酸菌、芽胞、真菌、およびウイルスの11種の異なる微生物のAmerican Tissue Culture Collection(ATCC)株に対する、本化合物の濃度別殺微生物活性を調べることである。米国環境保護局の基準(United States Environmental Protection Agency[EPA]Standards)に従い、毒性試験も実施した。室温で5分間の接触後に殺微生物活性を示した過酸化2-ブタノン濃度は、レジオネラ・ニューモフィラ(Legionella pneumophila)に対しては0.12%、大腸菌、緑膿菌、およびエンテロコッカス・ヒラエ(Enterococcus hirae)0.5%、黄色ブドウ球菌1%であった。抗酸菌に対する殺菌活性は0.5%の濃度で20℃、60分間接触後に、芽胞に対する殺菌活性は4%で40℃、60分間接触後に得られた。真菌(酵母、糸状菌)およびウイルス(アデノウイルス、ポリオウイルス)に対する十分な殺微生物活性は、0.5%で60分間接触後に得られた。毒性評価については、急性皮膚刺激試験、急性眼刺激試験、および急性経口毒性試験の結果は陰性であった。皮膚感作試験は陰性であった。毒性試験での安全性プロファイル、および試験株に対する基本的な殺微生物活性から、病院感染管理に過酸化2-ブタノンを使用することが推奨される。

監訳者コメント:
新規化学物質の検証を行う際には、消毒薬・殺菌剤としての役割以外にも数多くの検討がなされるべきである。新規化学物質の導入はかなり慎重に検討する必要がある。一方で、国内ではこうした新規化学物質の導入にあまりに神経質なため、欧米諸国とのギャップも大きくなっている。
