乾燥表面バイオフィルムからの微生物の移動と黄色ブドウ球菌(Staphylococcus aureus)の低栄養および低湿度条件下での長期生存が病原性に及ぼす影響★

背景
乾燥した病院表面のバイオフィルムは、乾燥した厳しい臨床表面での微生物の存続を促進し、汚染された表面で感染性病原体のリザーバとして働く可能性がある。
目的
本研究では、乾燥バイオフィルムからの接触を介する生存黄色ブドウ球菌(Staphylococcus aureus)細胞の移動を定量し、栄養および湿度不足が黄色ブドウ球菌の病原性レベルに及ぼす影響を調べた。
方法
臨床表面での乾燥バイオフィルム形成を模倣する最適化された条件下で、24 ウェルプレートにおいて、黄色ブドウ球菌 ATCC 25923 とバイオフィルム形成能を欠損した変異株である黄色ブドウ球菌 1132 の乾燥バイオフィルムを形成させた。乾燥バイオフィルムの接触を介して微生物細胞の移動を誘導し、栄養寒天上で定量した。栄養および湿度の不足が病原性レベルに及ぼす影響を調べるために、黄色ブドウ球菌 ATCC 25923 の乾燥および標準バイオフィルムならびに浮遊細胞を Galleria mellonella に接種し、殺傷率を比較した。
結果
本研究の結果から、黄色ブドウ球菌 ATCC 25923 の乾燥バイオフィルムに由来する生存細胞は、病原性が有意に高く、接触試験を介して乾燥バイオフィルムから容易に移動可能であり、したがって感染リスクがより大きいことが示された。黄色ブドウ球菌株のバイオフィルム形成能は移動可能性に有意な影響を及ぼさず、バイオフィルム表面が湿潤しているときにより多くの細胞が移動した。
結論
これらの結果から、病院表面の乾燥バイオフィルムは、病院内の病原性微生物伝播のリザーバとなる可能性があることが示されており、したがって病院表面の定期的な清掃と適切な消毒の重要性が強調される。
