医療における汚染除去剤として可能性のあるプラズマ活性溶液:抗菌活性および清掃用クロス使用の評価★

背景
低温プラズマは、抗菌特性を有する高濃度の活性酸素種および活性窒素種(例えば、亜硝酸塩)によりプラズマ活性溶液を発生させる。
目的
プラズマ照射生理食塩水から生成されたプラズマ活性溶液中の活性酸素種および活性窒素種の濃度を調査することと、プラズマ活性溶液による浮遊性細菌の不活化および汚染された清掃用クロスの除染を評価することを目的とした。
方法
圧縮空気による低温プラズマ照射によりリン酸緩衝食塩水(PBS)を 90 秒または 300 秒処理し、プラズマ活性溶液を生成した。活性酸素種および活性窒素種を定量的蛍光アッセイ(2,7-ジクロロフルオレセイン二酢酸)と比色アッセイ(Greiss 法)を用いて定量した。メチシリン耐性黄色ブドウ球菌(methicillin-resistant Staphylococcus aureus[MRSA])および大腸菌(Escherichia coli)の分離株をプラズマ活性溶液中で一晩培養し、不活化をコロニー形成単位(cfu)アッセイにより評価した。清掃用クロスの切片を MRSA と大腸菌とともに培養し、プラズマ活性溶液で 1 時間処理した。レサズリン還元アッセイにより細菌の不活化を評価した。
結果
亜硝酸塩は、未処理の PBS 中で 0.1 μM であったのが、低温プラズマで 90 秒、300 秒処理したプラズマ活性溶液中では、それぞれ 49.1 μM、94.0 μM に増加した。活性酸素種は、未処理の PBS 中で 30 μM であったのが、低温プラズマで 90 秒、300 秒処理したプラズマ活性溶液中では、それぞれ 75 μM、103 μM に増加した。90 秒処理のプラズマ活性溶液は、MRSA および大腸菌の生存率を低下させ(P < 0.05)、300 秒処理のプラズマ活性溶液では、両細菌とも 7 log 超の減少となった。プラズマ活性溶液中の汚染された清掃用クロスの 1 時間処理は、生存可能な MRSA および大腸菌のそれぞれ 55%、73%の減少をもたらした(P < 0.05)。
結論
浮遊性細菌の不活化は、活性酸素種および活性窒素種の濃度と相関した。プラズマ活性溶液は、清掃用クロスを汚染した細菌を減少させた。この溶液は、清掃用クロスなどの院内消毒薬としての可能性がある。
