室内空気中の SARS-CoV-2 量を定量するためのプロトコールの効率と感度の最適化★

背景
感染症の予防・制御のために、空中微生物の定量法の開発が必要である。医療現場で報告される結果の相違が原因で、空気中の重症急性呼吸器症候群コロナウイルス-2(SARS-CoV-2)の RNA 量を評価する最も効率的で感度の高い戦略はどれかを結論づけるのは難しい。
目的
我々が以前に報告した SARS-CoV-2 RNA 量を測定するプロトコール(液体インピンジャーによるバイオエアロゾルの捕集とドロップレットデジタル PCR による RNA の定量に基づいたもの)を改善すること。
方法
COVID-19 患者の病室から空気サンプルを採取し、別々のエアーサンプラー、液状捕集媒体、逆転写酵素(RT)の効率および/または感度を評価すること。
結果
ミネラルオイルは完全性を損なわずに、親水性の媒体よりも良好に空中の RNA を保持する。BioSampler とミネラルオイルを用いて、空気サンプルの 80%で標的となる SARS-CoV-2 ORF1ab が検出された。ゼラチンメンブランフィルターを備えた MD8 sampler は有効性に有意差は認められなかったが、後者によって得られた空気 1 m3 あたりの SARS-CoV-2 コピー数は少なかった(28.4 ± 6.1 vs 9 ± 1.7)。SuperScript II RT は、単一 SARS-CoV-2 ゲノム RNA 分子をドロップレットデジタル PCR により高効率で検出することを可能にする。これは空気サンプルから標的となる SARS-CoV-2 N1 を検出することを可能にした唯一の RT であった。
結論
室内空気の SARS-CoV-2 RNA 量を定量するプロトコールの捕集効率と検出感度は、本研究で改善された。このような最適化は、室内空気の微生物学的安全性に関する我々の理解を深めるために重要である。
監訳者コメント:
SARS-CoV-2 の感染経路としてのエアロゾル感染は周知の事実であるが、空気中のウイルスの定量検出の方法に関しては標準化されていないのが現状である。空気中のウイルスの捕集方法と保存についても標準的な方法が定まっておらず今後のさらなる検討が必要である。
