酸化第一銅および酸化第二銅でコーティングされた表面の抗微生物活性★

背景
疾患は、汚染された表面(媒介物)との接触によって拡散しうる。例えば、媒介物は、メチシリン耐性黄色ブドウ球菌(meticillin-resistant Staphylococcus aureus;MRSA)や緑膿菌(Pseudomonas aeruginosa)の拡散と関係づけられている。抗微生物表面処理は、媒介物からの疾患伝播を低減する可能性のある方法であり、広域抗微生物活性が望ましい。
目的
酸化第一銅(Cu2O)および酸化第二銅(CuO)によるコーティングについて、12 の微生物(細菌および真菌を含む)に対する抗微生物活性について試験すること。
方法
2 つの表面コーティングを作製した。Cu2O は、ポリウレタンを用いた簡単な 2 段階プロセスにより、活性酸化銅粒子と結合させて作製した。CuO は空気中の Cu2O に対する加熱処理により調製して、CuO を産生させて早期焼結を引き起こすことで、継続的コーティングを形成させた。抗微生物活性について、微生物懸濁液飛沫 10μL により、細胞数をコロニー形成単位(cfu)として計数することで検討した。
結果
コーティングにより、9 種類の微生物、例えばグラム陰性およびグラム陽性菌、抗酸菌、および真菌などが速やかに殺滅された。例えば、Cu2O/ポリウレタン コーティングにより、1 時間後に 99.9997%の緑膿菌および 99.9993%の黄色ブドウ球菌が殺滅された。毎週の清掃後も、有効性は低下しなかった。Cu2O コーティングの抗微生物活性は、摩耗処理後も変化せず、コーティングにはヒト細胞に対する毒性はなかった。
結論
広域抗微生物活性、摩耗耐性、および Cu2O コーティングの低毒性が組み合わさることで、医療環境における使用可能性が示唆される。
監訳者コメント:
酸化第一銅(Cu2O)および酸化第二銅(CuO)によるステンレスへのコーティングについて、広域スペクトルの抗菌活性、耐摩耗性および低毒性を示した研究。医療現場での応用が期待されるものの、手指衛生などの標準予防策が行われている中で、黄色ブドウ球菌や緑膿菌などの伝播の抑制にどれほど寄与するかは評価が難しいところである。
