低濃度の過酸化水素蒸気の殺菌効果と BSL-3 実験室での検証

背景

バイオセーフティ実験室では感染性の高い病原体が培養され研究されている。実験室感染を防ぐため、これらの実験室を徹底的に消毒することは非常に重要である。過酸化水素を用いた、非接触であり腐食を軽減した部屋全体の消毒戦略を提示し評価した。

 

目的

実験室環境において、芽胞および細菌を生物指標として使用し、8% と 10%の過酸化水素蒸気(VHP)の殺菌効果を評価すること。

 

方法

バチルス・アトロファエウス(Bacillus atrophaeus)およびバチルス・ステアロサーモフィルス(Bacillus stearothermophilus)の芽胞を、細菌である大腸菌(Escherichia coli)、黄色ブドウ球菌(Staphylococcus aureus)、白色ブドウ球菌(Staphylococcus albus)とともに密閉された実験室の事前に設定された場所に置き、OXY-PHARM NOCOSPRAY2 VHP 発生装置を適用した。芽胞殺傷効果は定性的に評価し、殺菌効果は定量的に評価し、平均 log10 減少値を決定した。最後に、最適化した消毒戦略をバイオセーフティーレベル 3(BSL-3)の実験室で検証した。

 

結果

VHP 8% および 10%に 2 ~ 3 時間曝露した場合、選択した芽胞および細菌のそれぞれに関して微生物量の有意な減少が得られた。黄色ブドウ球菌は大腸菌および白色ブドウ球菌よりも耐性が高いことが分かった。VHP 8%への 3 時間を超える曝露の試験によって、BSL-3 実験室の表面および設備上のB. atrophaeusに対する 100%の殺傷率が示された。

 

結論

VHP 発生装置は拡散性が良好であり、腐食性が低く、消毒残留物の除去の時間を節約できる方法である。本研究は、様々な条件下のバイオセーフティ実験室の空気および物体表面の的確な消毒に関する参照情報をもたらすものである。

過酸化水素蒸気の殺菌効果は様々な条件で評価されている。本研究ではバイオセーフティレベル 3 の実験室での有効性を検証したものである。これらの消毒装置は主には病棟などで使用されることが多いが、このような検査室や、内視鏡室など体液汚染の多い部署での活用も有効かもしれない。
