本研究の目的は、バイオフィルム産生大腸菌(Escherichia coli)7株のバイオフィルム形成に対する、ヒト血清アルブミン、イブプロフェン、およびN-アセチル-L-システイン(NAC)の影響を評価することである。ポリスチレン製マイクロタイタープレートを用いた静置培養によりバイオフィルム形成を測定した。細菌増殖およびバイオフィルム形成に対する上記3成分の影響を、各成分を溶液として添加した場合、およびポリスチレン製ウェルへの前処理(コーティング)に使用した場合で調べた。溶液で測定した最小バイオフィルム形成阻止濃度は、ヒト血清アルブミン8 mg/L(全分離株)、イブプロフェン2~125 mg/L(5株)、およびN-アセチル-L-システイン30~125 mg/L(5株)であった。ポリスチレンプレートをヒト血清アルブミン8 mg/Lおよび32,000 mg/Lで前処理することにより、すべての分離株のバイオフィルム形成が有意に減少したのに対して、イブプロフェン125 mg/LおよびN-アセチル-L-システイン1,000 mg/Lによるコーティングでは有意な減少はみられなかった。前処理法でヒト血清アルブミン8 mg/Lおよび32,000 mg/Lと、イブプロフェン125 mg/LまたはN-アセチル-L-システイン1,000 mg/Lのいずれかを併用した場合、さらに強力なバイオフィルム抑制効果が複数の分離株で認められた。今回の結果から、ヒト血清アルブミン単独もしくはイブプロフェンまたはN-アセチル-L-システインのいずれかとの併用で医療器材をコーティングすることにより、大腸菌バイオフィルム形成が抑制される可能性が示唆された。さらに、イブプロフェンおよびN-アセチル-L-システインは、細菌増殖とバイオフィルム形成の両方に対する抑制効果があることから、大腸菌による尿路感染症の治療に有用である可能性がある。
