Quantitative measurement of the efficacy of protein removal by cleaning formulations; comparative evaluation of prion-directed cleaning chemistries

変異型クロイツフェルト・ヤコブ病の原因となる感染性因子は安定性が高いため、医原性クロイツフェルト・ヤコブ病の伝播の可能性がある場合は、その制御のための手術器具の洗浄が重要となる。本研究では、テストソイルに熱安定性アデニル酸キナーゼ（tAK）を加えてステンレススチール上に塗布し、この代替汚染物質に対する種々の化学洗浄剤の効果を、卓上洗浄消毒器を使用した場合、予浸のみの場合、および予浸後に卓上洗浄消毒器を使用した場合で評価した。まず2種類のtAKで除去の容易さの試験を行ったところ、スルホロブス・アシドカルダリウス（Sulfolobus acidocaldarius）由来のtAKのほうが残留性が強かったため、これを化学洗浄剤の評価に採用した。従来の化学洗浄剤は全般的に、卓上洗浄消毒器で使用するほうが予浸に用いるよりも効果が高かった。成績が中程度の酵素洗浄剤を使用した卓上洗浄消毒器による処理の前に予浸を行った場合は、洗浄効果が改善し、tAKの残存活性に対する効果は予浸のみと卓上洗浄消毒器洗浄との相加的効果よりも高かった。プリオンを標的とした4種類の化学洗浄剤のうちの3種類では、予浸単独でもtAK残存活性は4.8 log₁₀以上低下して定量下限値未満となり、残存tAKは175 pg未満であった。従来のアルカリ洗浄剤でもtAK残存活性は定量下限値未満に低下したが、これは卓上洗浄消毒器を使用した場合のみであった。ステンレススチール上の乾燥したtAKソイルは、湿潤なソイルと比較して効果的な除去が困難であり、予浸時と卓上洗浄消毒器処理時のtAK残存活性はそれぞれ320倍と28倍であった。本研究により、様々な化学洗浄剤または処理過程による蛋白質の除去効果を評価するうえで、tAKインジケータが有用である可能性が示された。
