モノクロラミンおよびクロラミンTの殺菌活性の比較

クロラミンT(CAT)およびモノクロラミン(NH2Cl)は活性塩素化合物であり、殺生物剤(バイオサイド)として知られている。クロラミンTは、分子量が小さく脂溶性が高いモノクロラミンより酸化活性が高い。これにより、酸化活性の低さを高い脂溶性によって補えるか否かが問題となる。この問題に対処するために、純モノクロラミンの抗菌活性および抗真菌活性をクロラミンTと比較検討した。20℃、pH 7.1の等モルのクロラミンTおよびモノクロラミン溶液中で、大腸菌(Escherichia coli)、黄色ブドウ球菌(Staphylococcus aureus)、緑膿菌(Pseudomonas aeruginosa)、アスペルギルス・フミガーツス(Aspergillus fumigatus)、アスペルギルス・フラーブス(A. flavus)、およびカンジダ・アルビカンス(Candida albicans)の定量的殺菌試験(killing assay)を行った。モノクロラミンは、全試験株に対してすべての試験濃度下でクロラミンTよりも優れていた。濃度0.036 mMでは、モノクロラミンは1分以内に大腸菌の菌数を、5分以内に黄色ブドウ球菌の菌数を、いずれも3 log10減少させたのに対し、クロラミンTは同じ効果を示すのにそれぞれ120分、30分を要した。濃度0.107 mMでは、モノクロラミンは5分後に緑膿菌を3 log10減少させたのに対し、クロラミンTは20分後であった。モノクロラミン0.355 mMは30秒以内にC. albicansを2 log10減少させたのに対し、クロラミンT 0.355 mMで同様の減少を得るためには60分を要した。両消毒薬の差は、アスペルギルス属を用いた試験でさらに顕著であった。モノクロラミンは酸化活性が低いにもかかわらず、クロラミンTよりも抗菌活性および抗真菌活性が有意に高かった。この現象は、その脂溶性および分子量が小さいことに起因する可能性があり、クロラミン含有消毒薬を開発する場合や使用する場合には考慮する必要がある。

監訳者コメント:
同じ活性塩素化合物系統でバイオサイドとしての性能比較をした論文である。脂溶性が高いということは、細胞内への浸透性が良いということを意味しており、これが有効性の差と関係しているのではないだろうか?
