Bacteriostasis testing on allograft tissue inoculated in Wilkinse-Chalgren broth

組織バンクでは、組織検体の抗菌薬による除菌後に、生菌がないことを確認するために培養検査を行う。そのため、除菌した組織に残留抗菌薬が付着し、濃縮培養液に混入する可能性がある。この場合、培養が阻害され、偽陰性となることがある。本研究の目的は、骨および腱の組織片を接種したWilkins-Chalgren培養液の静菌作用の測定である。組織処理の過程にある2種類の組織片を、ゲンタマイシン含有溶液に浸漬した。米国薬局方記載の静菌性試験法に従って、試験菌株としてゲンタマイシン感受性緑膿菌ATCC 15442株、黄色ブドウ球菌ATCC 6538株、枯草菌（Bacillus subtilis）ATCC 6633株、対照菌株としてゲンタマイシン耐性カンジダ・アルビカンス（Candida albicans）ATCC 90029株を使用した。培養液中の残留ゲンタマイシン濃度を測定し、ブドウ球菌懸濁液を接種したミューラーヒントン寒天培地にゲンタマイシン浸漬組織を置床した。試験菌株では53～75％に静菌作用が認められた。静菌作用は腱組織片（85％）のほうが骨組織片（28％）よりも有意に高率に認められた。残留ゲンタマイシン濃度が最も高かったのは、腱組織片を接種した培養液であった。
