グループの異なる消毒薬によるヒトアデノウイルスのヘキソン変性★

ヒトアデノウイルスは、消毒薬の広域スペクトルの殺ウイルス効果を調べるための代用ウイルスとして、しばしば使用される。しかし最近の研究で、このウイルス群の化学物質感受性は大きく異なり、モデルウイルスとして推奨できるのは血清型5および44のみであることが示されている。本研究では、血清型1、2、5、6、8のヘキソン蛋白質を消毒薬に曝露し、次いでウエスタンブロット法およびRPS Adeno Detectorによる検出を行った。60分以内にヘキソン蛋白質が完全に変性したのは、比較的濃度の高い(0.5%)過酢酸のみであった。この効果は検査を行ったすべてのアデノウイルスで一様であり、0.05~2.5%ポビドンヨード(PVP-I)または0.7%ホルムアルデヒドへの曝露では認められなかった。しかし、ウイルス感染価およびゲノムの完全性は、PVP-I、ホルムアルデヒド、および低濃度のPAAにより影響を受けた。結論として、ヒトアデノウイルスのヘキソン蛋白質の消毒薬に対する抵抗性は予想以上に高く、一様であった。アデノウイルスの化学物質感受性の差を、その主要な構造化合物の感受性で説明することはできないが、今回の知見は、消毒薬の殺ウイルス作用に関する新しい解釈を提供するものである。

アデノウイルスはエンベロープを持たず、252個のカプソメアよりなる正20面体構造をしている。そのうち頂点にある12個は突起構造を持ったペントン(ペントンベースとファイバーからなる)と呼ばれ、他の240個はヘキソンと呼ばれる。本論文は、ウイルスに対する消毒薬の効果を検定する上で具体的にどのウイルス株を用いるべきなのか、質的な差が異なる血清型により生ずるのかについて検討したものである。
