統合した細胞培養とリアルタイム RT-PCR を組み合わせた懸濁液中のエボラウイルスの不活化に対する消毒薬の有効性★

背景
エボラウイルスは、感染者の分泌物または排泄物で汚染された環境表面(媒介物)との接触を介して伝播し得る。公衆衛生上の緊急事態を引き起こし得るため、また有効な治療薬やワクチンがないため、重要な介入法として、汚染された表面を消毒するための効果的な殺ウイルス製剤の使用が考えられる。
目的
エボラウイルスに対する以下の 3 種類の消毒薬について殺ウイルス効果を評価すること:Micro-Chem Plus 洗浄用消毒クリーナー(MCP)、FWD(*訳者注:フォワード[消毒薬の名称])、およびエタノール。中国消毒技術基準に従った懸濁試験を実施した。
方法
すべての製剤を様々な濃度で、曝露時間の範囲を 15 秒から 8 分として、定量的懸濁試験を行い、不活化を比較する分析を行った。ウイルス力価が 4 log10 以上減少した場合、殺ウイルス活性を認める所見とした。
結果
2 種類の第 4 級アンモニウム化合物を含有する MCP と FWD は有効性が高く、エボラウイルスを接触時間 15 秒以内で不活化したが、低濃度で用いた場合は両者の間にわずかな差が認められた。文献の記述と一致して、本研究の結果から、医療用エタノールのエボラウイルスに対する優れた殺ウイルス活性が確認され、これにより濃度 38%(v/v)で 15 秒以内にウイルス力価をバックグラウンドレベルにまで減少させられる。
結論
これら 3 種類の消毒薬は、適切な短い接触時間でエボラウイルスに対する十分な不活化効果を示し、これは現場においても実用的に達成できると考えられる。医療環境および検査室の除菌のためにこれらの消毒薬を使用することで、エボラウイルス伝播のリスクを低減できるであろう。
監訳者コメント:
消毒薬の有効性は標的生物だけでなく、適用方法にも依存している。消毒薬も新製品が市場に登場してきているが、有効性を評価するために標準化された検査が必要である。本研究ではエボラウイルスに対する 2 つの第 4 級アンモニウム塩の消毒薬とエタノールの有効性を証明している。希釈後 1 週間後にも消毒薬の有効性を評価しており、1 週間後には有効性が大幅に低下している。消毒薬希釈後は長期に使用してはいけないことを改めて理解できる。
