背景
滅菌サービス部門での手術器具の汚染除去処理では、プリオン蛋白など強固な感染性汚染物質を効果的に除去しようと努力がなされている。近年デザインされた新規システムは、低圧の超音波活性化冷水流を使用しており、硬表面における複数の生物学的汚染物質の効果的な洗浄が既に実証されている。
目的
外科用ステンレススチール表面からプリオン関連アミロイドなど組織蛋白を除去するための超音波活性化水流の有効性を試験すること。
方法
22L、ME7、または 263K プリオン株感染の脳ホモジネートで試験用の表面を汚染させた。汚染させた表面を超音波活性化水流で接触時間 5 秒間および 10 秒間の処理を行った。残存する蛋白性物質およびアミロイドの汚染を高感度顕微鏡解析により定量化し、超音波活性化水流による処理前後のプリオンの溶出残渣を免疫ブロット法を用いて確認した。
結果
外科用ステンレススチール表面からの異なるプリオン株の効果的な除去が観察され、回収された上澄みにおいてプロテアーゼに感受性および耐性を示すプリオン蛋白の濃度の低下が認められた。
結論
本研究により、超音波活性化水流は、現行の汚染除去法を改善する費用対効果の高い解決策となりうること、そして、院内感染の減少につながる可能性があることが確認された。
