Potential use of targeted enzymatic agents in the treatment of Staphylococcus aureus biofilm-related infections

黄色ブドウ球菌（Staphylococcus aureus）は、医療関連感染症の主な原因である。黄色ブドウ球菌の植込み型医療器材表面および宿主組織に対する接着・蓄積能力は、この病原体が引き起こす感染症の治療を困難にしている。現行の治療は、表面に接着する黄色ブドウ球菌に対する効果に限界があることが示されているが、バイオフィルム分解酵素剤（以下分解剤）を追加することによって効果が増強される可能性がある。本研究は、メチシリン耐性およびメチシリン感受性の黄色ブドウ球菌株によって形成されたバイオフィルムに対する、dispersin B、リソスタフィン、α – アミラーゼ、V 8 プロテアーゼ、およびセラペプターゼの酵素製剤を単独またはバンコマイシンおよびリファンピシンとの併用下で評価した。どちらの抗菌薬も、いずれかの分解剤との併用下で効果が増強された。リソスタフィンおよびセラペプターゼが、調査したすべての菌株に対して最も有効な分解剤であった。これらのデータから、バイオフィルム分解剤と抗菌薬との併用によって、黄色ブドウ球菌バイオフィルム関連感染症の治療選択肢が増える可能性が示されている。

監訳者コメント：
黄色ブドウ球菌や表皮ブドウ球菌は、人工物表面にバイオフィルムを形成するため、バイオフィルム感染症を発症すると人工物の抜去以外に適切な治療方法がないのが現状である。本実験ではこれらのバイオフィルム構造を分解する酵素製剤と抗菌薬を併用することで相乗効果が in vitro において認められたことから、新たなバイオフィルム感染症治療への可能性を報告している。
