現在のオートクレーブ処理の目的は細菌の死滅である。オートクレーブ処理が細菌ゲノム DNA の完全性に及ぼす作用についてはほとんど知られていない。細菌 DNA の完全性の指標として PCR 法を使用して、標準的なオートクレーブ処理が細菌 DNA の完全性に及ぼす影響を評価する実験を行った。増幅可能な PCR シグナルが観察されるオートクレーブ処理時間は、緑膿菌(Pseudomonas aeruginosa) NCTC 10662 では 10 分、20 分、および 30 分、サルモネラ(Salmonella) Nottingham NCTC 7832 では 10 分、20 分、30 分、および 40 分、大腸菌(Escherichia coli) NCTC 9001 では 10 分および 20 分であった。このことから、死滅した細菌やより高等な微生物のゲノム DNA 遺残がリザーバとして働き、遺伝子水平伝播によって毒性、持続性、抗菌薬耐性などの性状をつかさどる危険な遺伝子が生菌に導入される可能性があるため、今後のリスク評価や環境への影響の評価の際は、このような分子的遺残について慎重に考慮する必要がある。
