感染性プリオン蛋白質（PrP^Sc）は、多くの化学的・酵素的汚染除去法に耐性を示すこと、およびステンレススチールに強力に接着する性質があることから、PrP^Sc の医原性伝播は潜在的な脅威である。術中・術後の外科用器具の取り扱い状態は、組織蛋白質やプリオンの接着量およびその後の汚染除去法の効果に影響を及ぼす可能性がある。本研究では、外科用ステンレススチールへの組織蛋白質およびプリオンアミロイドの接着を時間および種々の保存条件との関連で評価するとともに、その後の酵素洗浄剤の効果を調査した。ME7 感染脳ホモジネートで表面を汚染させ、乾燥または湿潤状態の室温で 0 ～ 120 分または 24 時間乾燥させた。高感度の蛍光染色法と落射型微分干渉/エピ蛍光顕微鏡（episcopic differential interference contrast/epifluorescence microscopy）を使用して、洗浄前後の残存汚染を可視化した。乾燥時間が長い場合は、蛋白質およびプリオンアミロイドのいずれも吸着レベルが増加し、評価した洗浄剤の効果に影響した。湿潤環境での汚染後は、外科用ステンレススチール表面への蛋白質およびプリオンアミロイドのいずれも接着レベルが有意に減少した。湿潤状態を維持することにより、再利用可能な手術器具のその後の汚染除去効果が改善し、処理時間および費用も減少すると考えられる。
