The development of a new three-step protocol to determine the efficacy of disinfectant wipes on surfaces contaminated with Staphylococcus aureus

消毒ワイプの有効性、微生物を除去し表面からの伝播を防止する能力、および全体的な抗菌活性を定量化するための3段階のプロトコールを開発した。有機物負荷がある状態とない状態で、メチシリン耐性黄色ブドウ球菌（MRSA）またはメチシリン感受性黄色ブドウ球菌（MSSA）（6～7 log10 cfu）をステンレススチールのディスクに接種した。試験用のグレープフルーツ抽出液含有ワイプと、医薬部外品の対照ワイプを用いた。第1段階では、ワイプをディスクの表面に当て、100±5 gの重量をかけながら機械的に60 rpmで10秒間回転させた。スチール製ディスクを中和剤に移して再度懸濁し、残存細菌数を算出することにより、細菌除去の評価を行った。第2段階では、ワイプを中和剤含有寒天プレートに機械を用いて連続8回圧着させ、ワイプ由来の微生物伝播を評価した。第3段階では、ワイプに10秒間、直接接種を行い、次に中和等の処置を施し、抗菌活性を評価した。試験ワイプは対照ワイプと比較して、すべての表面について細菌性細胞に対する有意に高い除去効果を示した（P＜0.05）。しかし、ワイプの殺菌活性は低く（直接接種で＜1 log10の減少）、処置後ワイプ上に細菌が残存したため、初期の汚染レベルが高い場合は微生物伝播が繰り返し生じた。MRSAとMSSAに、除去、伝播、または抗菌活性の差はみられなかった。この3段階の方法は、表面消毒用ワイプの性能の評価ガイドラインを開発するために有用なツールである。
