Electro-elution, a novel method to remove transmissible spongiform encephalopathy-associated PrPSc from stainless steel surgical instruments

手術器具を通して伝染性海綿状脳症の医原性伝播が生じることが明らかにされており、再使用する器具を処理するために使用される従来の汚染除去技術の有効性が懸念されている。本稿では、伝染性海綿状脳症に特異的な異常蛋白質であるPrPScをステンレス・スチール製手術器具の表面から除去するための新しい洗浄法である「電気的溶出（electro-elution）」の開発について述べる。電気的な溶出過程では、蛋白質沈着物を除去するために電解質緩衝液の存在下で、ステンレス・スチール製手術器具に電流をかける。ステンレス・スチール製のディスクを感染性脳ホモジネートで汚染し、電気溶出による付着物除去能力を判定するために、種々の条件下で処理した。また、電気溶出後のディスク上のPrPScの残存を判定するために、新しい検出法である「直接ブロッティング法（direct blotting）」を開発した。直接ブロッティング法では、検出にあたって、蛋白質の器具表面から蛋白質親和性膜への直接的な受身伝達プロセスを利用する。本研究は、電気溶出はステンレス・スチール製手術器具の表面から疾患関連PrPScを効果的に除去し、おそらくは分解する能力を有することを示している。
