Validation of biocides against duck hepatitis B virus as a surrogate virus for human hepatitis B virus

B型肝炎ウイルスに対する消毒薬の殺ウイルス作用検査のための代替ウイルスとして、アヒルB型肝炎ウイルス（DHBV）の使用が推奨されてきた。しかし、このモデルは検査室で使用するには相当な困難を伴うことから、現在では欧州諸国においては標準的な検査法とは見なされていない。これまでの研究が示すように、いくつかの代替手段により、細胞培養系でのDHBV感染の検証が改善すると考えられる。間接免疫蛍光法による抗原染色およびライトサイクラーリアルタイムPCR法を利用して、先天的に感染したアヒルから得たDHBVまたはトランスフェクトした肝癌D2細胞系から採取したDHBVに対する、過酢酸（PAA）、ポビドンヨード（PVP-I）、およびホルムアルデヒドの抗ウイルス作用を検査した。その結果から、D2細胞系由来DHBVの不活化は、低濃度の消毒薬および短時間の曝露で達成されることが示された。D2細胞系由来DHBVに対する濃度-曝露時間値が感染アヒル由来DHBVと比較して低いことは、D2細胞系由来ウイルスの感受性が高いことを示している。加えて、懸濁試験でDHBVを有意に不活化することができたPAA濃度およびPVP-I濃度によっても、ウイルスゲノムを破壊することはできなかった。結論として、DHBVに対する消毒薬の殺ウイルス作用の検査には、先天的に感染したアヒル由来のDHBVを使用するべきであり、D2細胞系由来DHBVは、消毒薬に対する感受性が高いため不適切である。間接免疫蛍光染色法によりDHBV感染性の信頼性の高い検出が可能であるが、一方、定量的PCR法により抗ヘパドナウイルス作用の評価が可能である。
