新抗菌薬OxsilR 320Nに対するバイオフィルム形成細菌および浮遊細菌の感受性

標準EN 1040およびNF T 72-150※により、浮遊細菌に対する消毒薬の有効濃度を判定した。これは接触5分後に殺菌効果をもたらす濃度であり、その後中和される。しかし微生物はしばしば下層に定着し、そこで細胞外ポリマー・マトリクスに包まれて微小コロニーやバイオフィルムを形成する。バイオフィルムは通常、多種類の抗菌薬に耐性を有するが、耐性の機序には、細胞外ポリマー・マトリクス、遺伝子発現の変化、代謝の変動が関係している。このように耐性機序が多様であるため、バイオフィルムに対する有効な抗菌薬の選択と濃度設定が困難である。こうした状況の下で、SODIFRA社は新規消毒薬OxsilR 320N(フランス特許94 15 193)を開発した。Oxsil 320Nは、過酸化水素、酢酸・過酢酸、銀の3種類の有効成分が混合したものである。浮遊細菌、およびステンレス鋼であるAISI 304の表面における形成後24時間のバイオフィルムに対して、本殺菌剤の試験を行った。また、SODIFRA社の推奨事項(殺菌剤の中和は行わない)に従って、バイオフィルムでOxsil 320Nの有効濃度を判定した。浮遊細菌で測定した抗菌薬の有効性は、バイオフィルムの存在下での有効性の確実な指標とはいえないことが示された。バイオフィルムをOxsil 320Nに曝露した場合に、微生物濃度を105倍に減少させるために必要な濃度は、細菌懸濁液に対する濃度(0.313% Oxsil 320N)の10倍であった。Oxsil 320Nの有効濃度として、3.13%が必要であった。
